Sac-TMT extends survival over chemotherapy in EGFR-mutant lung cancer

Patients with EGFR-mutant lung cancer who progressed on TKI therapy gained 8.5 months of median survival on sac-TMT versus platinum-doublet chemo: 25.9 months to 17.4. The figure comes from the final analysis of the Phase III OptiTROP-Lung04 trial, presented at the 2026 ESMO Congress in Madrid. It makes sac-TMT the first TROP2-targeting antibody-drug conjugate to show a survival benefit over chemotherapy in this patient group.

A trial pairing an antibody-drug conjugate with the immunotherapy pembrolizumab cut the risk of disease progression or death by 51% in patients with PD-L1-negative advanced lung cancer. The OptiTROP-Lung06 results were presented at the 2026 European Society for Medical Oncology (ESMO) congress in Madrid and published the same day in the New England Journal of Medicine.
