InventisBio's oral TYK2 inhibitor meets Phase III psoriasis endpoints

InventisBio's oral TYK2 inhibitor meets Phase III psoriasis endpoints

New Capabilities

Nomelcitinib cleared skin lesions in more than 70% of patients at 16 weeks, setting up a head-to-head test against the only approved TYK2 drug.

Overview

Updated 2 hours ago

InventisBio reported on Nov. 2, 2026 that its oral drug nomelcitinib met the main and secondary goals of a 391-patient Phase III trial in moderate-to-severe plaque psoriasis. More than 70% of patients on either of two doses reached PASI 90 at week 16, meaning their psoriasis severity score fell at least 90% from baseline. More than 80% reached a physician rating of clear or almost clear.

Nomelcitinib is a TYK2 inhibitor, a class designed to damp inflammation by blocking a narrower set of immune signals than older JAK drugs. An oral option with skin-clearance rates in the range biologics achieve would compete with injected treatments and with Bristol Myers Squibb's deucravacitinib, the only approved TYK2 inhibitor. InventisBio is already running a head-to-head Phase III against that drug.

Why it matters

An oral pill that clears psoriasis in most patients would add a needle-free option to injected biologics.

Key Indicators

>70%
Patients reaching PASI 90 at week 16
Both nomelcitinib dose groups cleared at least 90% of baseline psoriasis severity in more than 70% of patients, versus placebo (p<0.0001).
>80%
Patients rated clear or almost clear at week 16
The physician global assessment secondary goal was met in more than 80% of treated patients, versus placebo (p<0.0001).
391
Patients enrolled in the Phase III trial
Participants were randomized 1:1:1 to two nomelcitinib dose groups or placebo across multiple centers.
16 weeks
Time to primary endpoint
Significant separation from placebo was already visible by week 4 and widened through week 16.
~1,000x
Preclinical selectivity for TYK2 over JAK1
InventisBio says the drug binds the JH2 regulatory domain of TYK2, a design intended to spare broader JAK signaling.

Scenarios

1

China approves nomelcitinib for plaque psoriasis

Likely Resolves by End of 2027

Discussed by: InventisBio, which described the trial as registrational, and Chinese securities press

The Phase III study was designed as a registrational trial, meaning it is intended to support a marketing application in China. A filing and approval would make nomelcitinib the first homegrown TYK2 inhibitor cleared for psoriasis in the country. The company has not disclosed a filing date.

2

Head-to-head trial shows nomelcitinib beating deucravacitinib

Uncertain Resolves by Q2 2028

Discussed by: InventisBio, which says the comparator trial is progressing smoothly

InventisBio is running a second Phase III that uses deucravacitinib as the active control instead of placebo. A statistically significant win on skin clearance would give nomelcitinib a claim to best-in-class status among oral TYK2 drugs. A tie or loss would leave it as one of several similar options.

3

US Phase II leads to a US Phase III trial

Possible Resolves by Q2 2028

Discussed by: InventisBio, which lists a US Phase II psoriasis study among ongoing programs

InventisBio is running a Phase II psoriasis trial in the United States. Positive data there would be the step needed before a US Phase III and eventual application to the Food and Drug Administration. The company has not announced a timeline for that decision.

Historical Context

2 moments from history that rhyme with this story — and how they unfolded.

December 2024

D-2570 Phase II readout (2024)

InventisBio reported results from a 161-patient Phase II trial testing three doses of D-2570 against placebo. Between 70.7% and 77.5% of treated patients reached PASI 90 at week 12, compared with 5% on placebo, and up to 50% reached complete skin clearance.

Then

The data established nomelcitinib as a serious late-stage contender and set expectations for Phase III.

Now

The Phase III results announced in November 2026 confirmed roughly the same magnitude of skin clearance at week 16.

Why this matters now

The Phase III trial was designed to confirm whether the Phase II response rates held in a larger, registrational study. They did.

2025

Deucravacitinib becomes the TYK2 benchmark (as of 2025)

By mid-2025, Bristol Myers Squibb's deucravacitinib was the only approved TYK2 inhibitor. About 23 TYK2 inhibitors were in development worldwide, 12 of them allosteric binders like nomelcitinib.

Then

Newer TYK2 candidates had to be measured against an approved oral drug rather than placebo alone.

Now

InventisBio designed a separate Phase III trial using deucravacitinib as the active control.

Why this matters now

Nomelcitinib's commercial case depends on how it compares with the one TYK2 drug already on the market.
